This is Ruffalo's first time playing Bruce Banner, having won the role after negotiations with Edward Norton went bad. He's said that he considers the role "this generation's Hamlet" and is excited to actually play Hulk. Whereas other actors have been replaced by CGI version of the raging green monster, the Avengers producers will be using the Avatar kind of technology of putting sensors on the actor and animating according to his frame.
In Iron Man 2, Natasha Romanoff was revealed to be an undercover agent working for S.H.I.E.L.D. Whedon has said that she'll be the only female Avengers member.
As of the end of Iron Man 2, Tony Stark decided that while Iron Man was eligible for the new Avengers initiative, he himself wasn't ready. He told Agent Coulson that he would stay on as a consultant.
But maybe the enemy in the movie will prompt him to take a more active role?

As we saw in the end of Captain America: The First Avenger -- spoiler! -- Cap has a lot to adjust to, having been frozen for more than 60 years. I think I'm most interested in his arc in this movie, as he goes from being a government project/propaganda figure to one of many superheroes.
Thor may not be ready to be king of Asgard, but he'll continue to fight for Earth.
Clint Barton/Hawkeye is the hero about which we know the least, but we do know that Renner trained extensively to play the man known as "the world's greatest marksman."
And here we have the agent of S.H.I.E.L.D.: Nick Fury, who's appeared in nearly every movie's credits to offer the hero the chance to assemble; Maria Hill, a new addition to the team; and Phil Coulson, who's also been a mainstay in the movies as he's the liaison with each of the heroes.
